About
2,5-Hexanedione — the human neurotoxic metabolite of n-hexane. Well-established peripheral nervous system toxicant; not appropriate for cosmetic use.
Acetonylacetone (2,5-hexanedione, CAS 110-13-4) is the principal toxic metabolite of n-hexane, the established cause of peripheral neuropathy in occupationally exposed workers (shoe, furniture, and printing industries). The diketone crosslinks neurofilament proteins, producing axonal swellings and distal axonopathy. This is one of the best-characterized industrial neurotoxicants. GHS classifications include flammability, acute oral toxicity, and serious eye irritation. It has no cosmetic safety monograph supporting use in leave-on products, and its systemic toxicology profile makes it unsuitable regardless of the route.
