Ambrein is the primary alcohol constituent of ambergris, the biliary secretion of the sperm whale (Physeter catodon). It is a tricyclic sesquiterpene with a characteristic warm, marine, sweet odor used as a high-quality fragrance fixative. Natural ambergris is now largely replaced by synthetic equivalents or verified sustainable sources due to CITES restrictions on endangered species. The EU Cosmetics Regulation does not ban ambrein itself, but requires verification of sustainable sourcing under CITES Appendix I/II. Toxicologically, it shows low acute toxicity with some structural similarity to known fragrance sensitisers, placing it in the mid-60s. IFRA standards apply and sensitisation testing in human subjects has not shown major concerns.
